Purposive regional sampling strata formulations are often inadequately defined in the literature, or are not based on objective, replicable, and formal criteria. The drainage class technique of strata formulation meets these criteria, yet is simple to apply, generates mapped strata directly, and makes use of topographic information commonly available in the United States. Statistical tests indicate drainage class strata covary significantly with environmental and cultural variables in the middle Salt River valley, northeast Missouri. Therefore, the approach should be useful in many other areas where ecological variation is expressed along altitudinal gradients which project upslope from elevations of major stream courses, particularly when more “traditional” stratification techniques cannot be applied
